Product Specific Information

PA1-852 detects KAP-1 (Kruppel-associated box-associated protein 1) in human, mouse, and rat samples.
It has been validated for use in Western blot, ICC/IF, and IHC (P) applications.
By Western blot, this antibody detects an approximately 87 kDa protein corresponding to recombinant human KAP-1, as well as a smaller band believed to represent a degradation product.
In immunofluorescent assays, this antibody primarily reacts with mouse or rat samples and may show non-specific staining in HeLa cells.
The PA1-852 antigen is recombinant human KAP-1.


Target Information

The KRAB (Kruppel-associated box) domain, approximately 45 amino acids long, is a transcriptional repression domain found in many transcription factors.
Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes have been identified in the human genome.
These proteins repress transcription through interactions with KAP-1 (KRAB-associated protein 1), an 835-amino-acid protein containing a RING finger, B boxes, and a PHD finger.
KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1).
KAP-1 targets KRAZ1 to centromeric heterochromatin regions containing HP1α, contributing to transcriptional repression.
Mutant KAP-1 proteins that bind KRAB but not HP1 lead to random KRAZ1 distribution and strong transcriptional activation.
