Product Specific Information

The 104 monoclonal antibody reacts with mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, AKR. CD45.2 is expressed by all leukocytes in these strains.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Tested on mouse splenocytes; ≤ 1.0 µg/test recommended
  • A test is defined as the amount (µg) of antibody that stains a cell sample in 100 µL.
  • Cell number may vary (10⁵–10⁸ cells/test). Optimal titration is recommended.

Fluorochrome Information:
Brilliant Ultra Violet™ 395 (BUV395) emits at 395 nm and is suitable for cytometers equipped with a 355 nm UV laser. Verify instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use:

Target Information

CD45.2 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, playing key roles in lymphocyte development and antigen signaling.
CD45.2 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. Isoform expression varies by cell type and physiological state.
Beyond immunoreceptor signaling, CD45.2 also promotes cell survival via integrin-mediated signaling and is involved in DNA fragmentation during apoptosis. CD45RA is an isoform with restricted expression among lymphoid subtypes.
