Product Specific Information

The WKH203 monoclonal antibody reacts with rat CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
While CD28 acts as a costimulator, CTLA-4 inhibits T cell activation by reducing IL-2 secretion and proliferation. The cytoplasmic region of CTLA-4 includes ER retention motifs, leading to intracellular localization of newly synthesized CTLA-4 upon TCR signaling.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on unstimulated and Con A-stimulated (3 days) rat splenocytes. Recommended ≤1 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Due to intracellular localization of CTLA-4, both surface and intracellular expression may need to be assessed for complete detection.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed mainly on CD4⁺ T cells. It binds CD80 and CD86 with higher affinity than CD28, delivering inhibitory signals to T cells. Intracellular CTLA4 is found in regulatory T cells and contributes to their suppressive function.
