Product Specific Information

The AA4.1 monoclonal antibody reacts with mouse C1qRp, an approximately 130–140 kDa C-type lectin-like type I transmembrane protein. The AA4.1 antigen was originally identified as an antigen expressed on early stages of B cell development in bone marrow. Adult spleen and fetal liver also contain detectable numbers of AA4.1⁺ cells.

When used with other hematopoietic progenitor markers such as Thy-1, Sca-1, c-Kit, CD43, and CD24, AA4.1 helps distinguish primitive and differentiated subsets of bone marrow lymphoid-committed progenitors (CLP). Co-expression of surface IgM (sIgM), CD23, and AA4.1 defines three nonproliferative subpopulations of immature or transitional B cells:

  • T1: AA4.1⁺ / CD23⁻ / sIgM^hi
  • T2: AA4.1⁺ / CD23⁺ / sIgM^hi
  • T3: AA4.1⁺ / CD23⁺ / sIgM^lo

AA4.1 is also expressed in cytoplasmic vesicles of endothelial cells, megakaryoblasts, and platelets. Monoclonal antibodies AA4.1 and 493 recognize different epitopes of the same molecule.

Applications Tested: Tested on mouse bone marrow and splenocyte cells; ≤0.125 µg per test recommended.
A test is defined as the antibody amount to stain a cell sample in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Optimize titration for best results.

Target Information

The encoded protein (CD93/C1qRp) is a cell-surface glycoprotein and type I membrane protein, initially identified as a myeloid cell-specific marker. Although once thought to be a receptor for C1q, it is now believed to function in intercellular adhesion and clearance of apoptotic cells. Its cytoplasmic tail interacts with moesin, linking the protein to the cytoskeleton and contributing to cytoskeletal remodeling.
