Target Information
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, undergoes a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA. This binding regulates gene transcription through interactions with coactivator or corepressor proteins.
Two main forms exist — ER alpha (ESR1) and ER beta (ESR2) — encoded by separate genes. Multiple isoforms of ER alpha are produced via alternative RNA splicing (66 kDa, 53 kDa, 47 kDa, 35 kDa). Estrogen receptors are widely expressed in various tissues, essential for sexual development, reproduction, and bone physiology, and are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
