Product Specific Information
The 4H11 monoclonal antibody reacts with human CD34, also known as mucosialin. CD34 belongs to a protein family that includes endoglycan and podocalyxin. These are single-pass transmembrane proteins with a heavily glycosylated extracellular mucin domain.
CD34 was first identified as an antigen expressed on hematopoietic progenitors and is widely used as a marker for isolating cells capable of hematopoietic engraftment. CD34 is also expressed on mesenchymal stem cells, tumor cell lines, and vascular endothelia.
Epitopes of CD34 are classified into three classes (I, II, III) based on enzymatic cleavage sensitivity. The 4H11 antibody belongs to class III, indicating it reacts with a protein epitope.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test in a final volume of 100 µL.
Cell number range: 10⁵–10⁸ cells/test.
Light Sensitivity: Protect from light; tandem dye is photo-sensitive.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Excitation: 488–561 nm
Emission: 667 nm

Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on many stem cell populations. It serves as a marker for hematopoietic stem cells and may function as a receptor regulating adhesion, differentiation, and proliferation.
CD34 expression reflects a specific hematopoietic developmental state and may mediate attachment to the bone marrow extracellular matrix or stromal cells. It may also act as a scaffold for lineage-specific glycans, facilitating interactions with lectins or selectins.
The intracellular domain of CD34 is phosphorylated by protein kinase C, suggesting a role in signal transduction.
Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
