Product Specific Information

The V3NTY24 monoclonal antibody recognizes human and mouse eukaryotic translation initiation factor eIF4E-binding protein 1 (4E-BP1) when phosphorylated at threonine 37 and/or threonine 46.
4E-BP1 acts as a translation repressor by binding to eIF4E in its non-phosphorylated form. Phosphorylation at multiple sites releases this repression, enabling cap-dependent translation.
FRAP/mTOR phosphorylates Thr37 and Thr46, while ATM phosphorylates Ser111. Phosphorylation at Thr37/Thr46 is inhibited by PI3 kinase inhibitors LY294002 and wortmannin.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells. Recommended use: 5 µL (0.06 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Recommended Protocols:

  • Protocol A: Two-step protocol for intracellular (cytoplasmic) proteins – optimal flexibility.
  • Protocol C: Two-step protocol (Fixation/Methanol) – best for phospho-specific discrimination.
  • Protocol B: One-step protocol for intracellular (nuclear) proteins – not recommended.

Target Information

This gene encodes a translation repressor protein that binds eukaryotic translation initiation factor 4E (eIF4E), inhibiting complex formation and translation initiation. Phosphorylation in response to stimuli such as UV or insulin signaling causes dissociation from eIF4E and activation of mRNA translation.
