Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reaction. The SK3 and RPA-T4 monoclonal antibodies recognize distinct epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.1 µg)/test in a final volume of 100 µL.
Cell number range: 10⁵–10⁸ cells/test.

NovaFluor Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.

CellBlox Plus Blocking Buffer:
This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Add 5 µL per sample for optimal staining.
Use 5 µL per 100 µL cell sample (10³–10⁸ cells) for single-color controls.

NovaFluor Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures for optimized fluorescence performance, minimizing spillover and spread.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Expressed mainly in T helper cells, it is also found in monocytes, macrophages, and dendritic cells. CD4 is detected in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. It contributes to T-cell activation and may mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
