Target Information
The expression of DUSP1 gene is induced in human skin fibroblasts by oxidative or heat stress and growth factors. It encodes a protein with structural features similar to non-receptor-type protein-tyrosine phosphatases and shares sequence similarity with a Tyr/Ser-protein phosphatase encoded by the vaccinia virus H1 gene. The bacterially expressed DUSP1 protein has intrinsic phosphatase activity, specifically inactivating MAP kinase in vitro by dephosphorylation of both phosphothreonine and phosphotyrosine residues. It also suppresses MAP kinase activation by oncogenic Ras in Xenopus oocyte extracts, suggesting a role in stress response and negative regulation of cellular proliferation.
