Target Information
Furin is a membrane-associated, calcium-dependent serine protease belonging to the subtilisin-like prohormone convertase (PC) family. These enzymes cleave prohormones, neuropeptide precursors, and various cellular, viral, and bacterial proteins transported via the constitutive secretory pathway.
Furin shares structural similarities with other PC family members, including:
- ~10 kDa amino-terminal proregion
- ~55 kDa conserved subtilisin-like catalytic domain
- Variable carboxyl-terminal domain
Furin becomes catalytically active after proregion cleavage within the appropriate compartment and is the only PC known to possess a transmembrane domain. It cleaves target proteins at the carboxyl-terminus of the consensus sequence RX(K/R)R. The acidic peptide sequence C771PSDSEEDEG780 localizes furin to the trans-Golgi network, and phosphorylation of serine residues modulates intracellular routing. The Y759KGL762 tetrapeptide sequence directs internalization from the cell surface.
