- Cellular Location: Nucleus and cytoplasm. Cytoplasmic in absence of ligand; migrates to nucleus when complexed with R-SMAD.
Target Information
Smad4 is encoded by the Smad4 gene located on chromosome 18 in humans. Also known as Smad family member 4, Sma- and Mad-related protein 4, or Mothers against decapentaplegic homolog 4.
Smad4 contains N-terminal MH1 and C-terminal MH2 domains involved in DNA binding and protein interactions.
Binding of TGF-beta or BMP ligands to their receptors leads to phosphorylation of Smad 1, 2, 3, 5, and 8 (R-Smad), which heterotrimerize with Smad4 (co-Smad) and translocate to the nucleus.
Inhibitory Smads (Smad6, Smad7) interact with activated R-Smads to attenuate signaling.
Smad4 functions as a tumor suppressor by transcriptionally regulating target genes such as Cyclin D1 (downregulation) and collagen (upregulation), thereby inhibiting cell proliferation.
Dephosphorylation regulates nuclear export and nucleocytoplasmic dynamics of Smads.
