Target Information
SMADs are members of the MAD-related family of intracellular proteins that play essential roles in the signaling pathways of serine/threonine kinase receptors within the transforming growth factor beta (TGF-β) superfamily.
They are categorized as:
- Receptor-regulated SMADs (R-SMADs): SMAD1, 2, 5, 8, and 9
- Common-mediator SMAD (Co-SMAD): SMAD4
- Inhibitory SMADs (I-SMADs): SMAD6 and 7
SMAD1, 5, 8, and 9 share high sequence homology, and antibodies often recognize common regions among them. SMAD8 and SMAD9 are frequently used interchangeably in literature. Mutations in SMAD9 are associated with pulmonary hypertension, primary 2 (PPH2).
