Target Information
Constant region of T cell receptor (TR) alpha chain (PubMed:24600447).
Alpha-beta T cell receptors are antigen-specific receptors essential to the immune response and are present on the surface of T lymphocytes. They recognize peptide–major histocompatibility (pMH) complexes displayed by antigen-presenting cells (APC), initiating TR-CD3 clustering and activation of LCK, which phosphorylates ITAM motifs of CD3G, CD3D, CD3E, and CD247 to recruit ZAP70.
ZAP70 phosphorylates LAT, leading to formation of the LAT signalosome that propagates signaling through calcium, MAPK, and NF-kB pathways, activating transcription factors crucial for T cell growth and differentiation (PubMed:9382891, PubMed:23524462).
The T cell repertoire is generated in the thymus through V-(D)-J rearrangement and shaped by selection events to produce self-MH restricted, non-autoaggressive T cells. Post-thymic interactions with pMH complexes further refine TR structural and functional avidity (PubMed:15040585). [UniProt]
