Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

The Wilms Tumor 1 (WT1) gene encodes one of the WPP domain-interacting proteins required for RanGAP nuclear envelope association in root tip cells.
Ran GTPase plays essential roles in nucleocytoplasmic transport, spindle formation, and postmitotic nuclear envelope reassembly.
The cytoplasmic RanGAP is critical for establishing a functional RanGTP/RanGDP gradient across the nuclear envelope and associates with the outer nuclear surface in metazoan and higher plant cells.
Arabidopsis thaliana RanGAP association with the root tip nuclear envelope requires plant-specific nucleoporins combining coiled-coil and transmembrane domains (CC-TMD) and WPP domain-interacting proteins (WIPs).
Wilms Tumor 1 and WIT2 have been identified as a second family of CC-TMD proteins, structurally similar yet distinct from the WIP family, required for RanGAP nuclear envelope association in root tip cells.
