Target Information

Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins linking the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a homology region that mediates binding to cell receptors and other TRAFs, activating various signal cascades depending on the TRAFs involved.
For example, TRAF2 and TRAF3 bind to CD40, a member of the NF receptor superfamily that induces B cell immunity, and are critical for NF-kappa-B activation in mouse B lymphocytes. TRAF2, along with TRAF6, is required for CD40 signaling in non-hematopoietic cells. TRAF2 also interacts with lymphotoxin-beta receptor (LTbetaR) with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
