Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, playing key roles in regulating cell death and stress responses.
TRAF proteins share a conserved region enabling receptor and TRAF–TRAF interactions, activating various signaling cascades.
For instance:

• TRAF2 and TRAF3 bind CD40, crucial for NF-κB activation in B lymphocytes.
• TRAF2 and TRAF6 are required for CD40 signaling in non-hematopoietic cells.
• TRAF2 also interacts with LTβR in association with TRAF3, cIAP1, and Smac.
