Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a marker for epigenetic transcriptional activation, thereby functioning as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 is part of several histone deacetylase complexes and silences genes via histone demethylation. It oxidizes substrates using FAD to generate imine intermediates that are hydrolyzed. LSD1 demethylates mono- and tri-methylated Lys-4 of histone H3 and may repress neuronal genes. It requires RCOR1/CoREST for nucleosomal activity and may also demethylate Lys-9 of histone H3, affecting androgen receptor target gene expression. Mutations in this gene can cause cleft palate, psychomotor retardation, and distinctive facial features.
