Target Information
PTEN은 종양 억제 인자로서 다양한 암에서 높은 빈도로 변이된 것으로 알려져 있습니다. PTEN 유전자는 phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase를 암호화하며, tensin 유사 도메인과 dual specificity protein tyrosine phosphatase와 유사한 촉매 도메인을 포함합니다. 이 단백질은 phosphoinositide 기질을 탈인산화하여 세포 내 phosphatidylinositol-3,4,5-trisphosphate 수준을 음성 조절하며, AKT/PKB 신호 경로를 억제함으로써 종양 억제 기능을 수행합니다. 또한 PTEN은 성체 신경발생 과정에서 신경세포의 위치, 수상돌기 발달 및 시냅스 형성 조절에 관여하며, 지방 조직에서 인슐린 신호 및 포도당 대사의 음성 조절자로 작용할 수 있습니다. 핵 내 단일 유비퀴틴화된 형태는 더 높은 세포사멸 가능성을 가지며, 세포질 내 비유비퀴틴화된 형태는 상대적으로 낮은 종양 억제 능력을 나타냅니다.
