Product Specific Information
Description: Interleukin-10, originally known as Cytokine Synthesis Inhibitory Factor (CSIF), is an 18.6 kDa protein of 160 amino acid residues that shares over 80% sequence homology with the Epstein-Barr Virus protein BCRFI. The reported biological activities of IL-10, which maybe interrelated, include inhibition of macrophage-mediated cytokine synthesis, suppression of the delayed-type hyper-sensitivity response, and stimulation of the Th2 cell response which results in elevated antibody production.
Source: Interleukin-10 is produced in E. coli.
Purity: Greater than 95% by SDS-PAGE and HPLC analyses.
Reconstitution: We recommend a quick spin followed by reconstitution in sterile destilled water to a concentration not less than 100µg/mL. This solution can then be diluted into other aqueous buffers and stored at 4°C for 1 week or -20°C for future use. For most in vitro applications, IL-10 exerts its biological activity in the concentration range of 0.2 to 20.0 ng/mL.
Target Information
IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes. This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice have suggested that this cytokine is an essential immunoregulator in the intestinal tract. Mutations in this gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
