Target Information
TNF alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers of two, three, or five noncovalently linked units, showing a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest neuroprotective functions and tumor necrosis induction in tumor-bearing mice.
TNF alpha also plays roles in immune responses to bacterial, viral, parasitic, and fungal infections and in tumor necrosis. It causes cytolysis or cytostasis of certain transformed cells, synergistic with interferon-gamma. TNF alpha has been implicated in autoimmune diseases, insulin resistance, and cancer.
