Product Specific Information
- Paraffin sections require pre-treatment prior to staining.
- Proteinase K is recommended for tissues fixed <24 hrs.
- Citrate buffer (pH 6.0) recommended for tissues fixed >24 hrs.
- For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
- Rat anti-mouse F4/80 antibody, clone Cl:A3-1 recognizes murine F4/80 antigen (~160 kDa cell surface glycoprotein), a member of the EGF-TM7 family.
- Shares 68% amino acid identity with human EMR1 (EGF module-containing mucin-like hormone receptor 1).
Target Information
F4/80 antigen is a 160 kDa cell surface glycoprotein belonging to the EGF-TM7 family. It shares 68% identity with human EMR1. Expression varies during macrophage maturation and activation.
F4/80 is expressed on mature tissue macrophages including:
- Kupffer cells
- Langerhans cells
- Microglia
- Macrophages in gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma, and spleen red pulp
It is also found on some dendritic cells but absent from macrophages in T cell areas of spleen and lymph node.
Recent studies suggest a role for F4/80 in generating efferent CD8⁺ regulatory T cells.
