MA3-912 항체는 개, 양서류, 인간, 생쥐, 쥐, 토끼 조직의 근소포체 또는 소포체 내 칼슘 펌프 SERCA1 ATPase를 검출합니다.
Western blot 및 면역형광(ICC/IF) 분석에서 성공적으로 사용되었으며, Western blot에서는 생쥐 근육 추출물 내에서 약 110 kDa 크기의 SERCA1 ATPase 단백질을 검출합니다.
개 골격근의 면역형광 염색 시, MA3-912은 빠른 수축형(type II) 근섬유 전체에 강한 신호를 보입니다.
또한 MA3-912은 바나데이트에 의해 유도된 SERCA ATPase 결정화를 억제하는 것으로 보고되었습니다.
이 항체의 항원은 정제된 토끼 골격근 근소포체이며, 토끼 SERCA1 ATPase의 아미노산 506번부터 C-말단까지 노출된 영역을 인식합니다.
Target Information
ATP 의존성 칼슘 펌프는 세포질 내 자유 칼슘 농도를 낮게 유지하는 데 기여합니다.
이 펌프들은 근소포체 또는 소포체 내에 존재하며, SERCA ATPase라 불리는 구조적으로 유사한 효소군에 의해 암호화됩니다.
- SERCA1: 빠른 수축형(type II) 골격근에서만 발현
- SERCA2: 조직 특이적 처리에 의해 SERCA2a(근육 특이형)와 SERCA2b(모든 세포형)로 나뉨
- SERCA3: 비근육 세포에서 발현되며, 상대적으로 덜 연구됨
