Product Specific Information

Description:
The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone, but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (common γ chain). The PC61.5 antibody inhibits IL-2 binding to both receptor classes, playing a role in lymphocyte differentiation and activation/proliferation.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and Con A-stimulated splenocyte culture. Recommended ≤0.125 µg/test in 100 µL volume. Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness. Performance may vary by clone and should be verified empirically.

Target Information

CD25 (IL2 receptor alpha chain, IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation. IL2RA forms a high-affinity receptor complex with IL2RB and IL2RG. Homodimeric IL2RA has low affinity, while IL2RB homodimers show medium affinity. IL2 is primarily produced by mature T cells and functions as a growth and differentiation factor, regulating cell death. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysregulation.
