Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells and is upregulated upon T-cell stimulation. CD27 binds to CD70, playing an important role in T cell–B cell interaction.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.25 µg) per test (final volume 100 µL). Cell number may vary from 10⁵ to 10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30. It regulates cell proliferation and death and is primarily expressed on lymphocytes. CD27 forms a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subpopulation of B cells. Activation via TCR-CD3 complex increases CD27 expression and release of soluble CD27 (sCD27), a prognostic marker for B cell malignancies. CD27-binding protein (SIVA) is involved in CD27-mediated apoptosis. Diseases associated with CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
