Product Specific Information

The JES3-9D7 monoclonal antibody reacts with human interleukin-10 (IL-10).
Applications reported include flow cytometric analysis and intracellular staining.
This antibody has been pre-titrated and tested by intracellular staining and flow cytometry analysis of Th2-polarized and stimulated human peripheral blood cells.
Recommended use: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody sufficient for staining a sample in a final volume of 100 µL. Cell number can range from 10⁵ to 10⁸ cells/test.

eFluor 660 is a replacement for Alexa Fluor 647, emits at 659 nm, and is excited with the red laser (633 nm). Ensure your instrument supports this fluorochrome.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and some lymphocytes. It plays a key role in immunoregulation and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It promotes B cell survival, proliferation, and antibody production, and can inhibit NF-kappa B activity. IL-10 is also involved in the JAK-STAT signaling pathway. Knockout studies in mice indicate its importance in intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and HIV Type 1.
