Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, together with other CD3 subunits (gamma and delta), is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed developmentally in thymocytes and on all mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes, recommended ≤1.0 µg/test (in 100 µL final volume). Optimize antibody titration per assay.
Fluorochrome Information:
Super Bright 436 is excited by the 405 nm violet laser and emits at 436 nm. Recommended detection filter: 450/50 bandpass (or equivalent). Ensure instrument compatibility.
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes subunits CD3γ, CD3δ, CD3ε, and CD3ζ, encoded by genes on chromosome 11. These subunits have cytoplasmic tails associated with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Reduced T lymphocytes may be observed in some autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
