Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by a majority of thymocytes, mature T cells, and a subset of B cells. CD5 expression on a small subset of B cells characterizes B-1 cells, a distinct lineage involved in immune regulation. CD5 serves as a counter-receptor for CD72 and plays a role in T-B cell interactions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Optimal performance requires empirical titration (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence signatures, minimizing spillover and spread.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein found on mature T-lymphocytes, thymocytes, and some B cells. It regulates T-cell proliferation and is expressed in most T-cell leukemias and lymphomas.

• Found on 95% of thymocytes and 72% of peripheral blood lymphocytes.
• Major reactivity in T-cell areas of lymph nodes.
• Associated diseases: thymus cancer, Richter’s Syndrome.
