Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. Expression of CD5 on a subset of B cells defines B-1 cells, which are developmentally distinct. CD5 serves as a counter-receptor for CD72 and plays a role in T-B cell interactions.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Cell numbers may range from 10⁵ to 10⁸ cells/test. Antibody titration is recommended for optimal performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) improves staining performance. Add 5 µL per sample to antibody cocktails/master mixes before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein expressed on mature T-lymphocytes, thymocytes, and many T-cell leukemias/lymphomas. It also reacts with a subset of activated B cells and may regulate T-cell proliferation. CD5 is found on 95% of thymocytes and 72% of peripheral blood lymphocytes, predominantly in T cell areas of lymph nodes. Associated diseases include thymus cancer and Richter’s Syndrome.
