Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells (B-1 cells). CD5 acts as a counter-receptor for CD72 and plays a role in T-B cell interactions.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes, ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL single-color control.
• NovaFluor conjugates are based on Phiton™ technology using nucleic acid dye structures for engineered fluorescent signatures and minimized spillover.

Excitation: 552 nm
Emission: 612 nm
Laser: 561 nm (Yellow)

Note: NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains. Other strains not tested.

Target Information

CD5 is a 67 kDa transmembrane glycoprotein expressed on most human thymocytes, mature T-lymphocytes, and certain B-cell subsets. It plays a role in T-cell proliferation regulation and is associated with diseases such as thymus cancer and Richter's Syndrome.
