Product Specific Information

The 53-7.3 monoclonal antibody reacts with mouse CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells (B-1 cells). CD5 is a counter-receptor for CD72 and plays a role in T-B cell interaction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometry on mouse splenocytes at ≤ 0.2 µg/test (100 µL volume, cell count 10⁵–10⁸ cells/test). Titrate for optimal performance.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures engineered for optimized fluorescence signatures with minimal spillover and spread.

Target Information

CD5 is a 67 kDa transmembrane glycoprotein found on mature T-lymphocytes, thymocytes, and certain B-cell subsets. It regulates T-cell proliferation and is expressed in T-cell leukemias, lymphomas, and activated T cells.
Associated diseases include thymus cancer and Richter’s Syndrome.
