Description:
The CD28.2 recombinant mouse monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and acts as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

The CD28.2 antibody has been reported for use in flow cytometric analysis and T cell costimulation assays in vitro.
Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells at 0.025 µg/test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be empirically determined (typically 10^5–10^8 cells/test). Antibody titration is recommended for optimal performance.
Target Information
CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein and a member of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and acts as a critical costimulatory receptor providing the second activation signal by binding CD80 and CD86 on antigen-presenting cells. CD28 prevents T cells from entering an anergic state or undergoing premature apoptosis. In murine lymphoid organs and blood, all CD4+ and CD8+ T cells express CD28, with expression patterns varying across thymocyte maturation stages. CD28 dysfunction has been associated with diseases such as mycosis fungoides and Sézary’s disease.
