Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment.

PD-L2 expression is upregulated by Th2 cytokine IL-4 and to a lesser degree by Th1 cytokines TNF-α and IFN-γ. Although generally expressed at lower levels than PD-L1, PD-L2 exhibits a 2–6 times higher affinity to PD-1. PD-1 and its ligands serve as inhibitory immune checkpoints, regulating T cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10), thereby maintaining immune homeostasis.

Interestingly, PD-L2 can also stimulate T cell proliferation and cytokine production even in PD-1-deficient cells, suggesting additional receptors such as repulsive guidance molecule b (RGMb). The PD-L2–RGMb interaction influences respiratory tolerance development in the lung via BMP and/or neogenin signaling pathways.

The human PD-L2 monomer comprises a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
