Target Information
Bassoon은 약 420 kDa 크기의 단백질로 시냅스 전 신경 말단에 위치하며, 시냅스 소포 순환의 구조적 및 기능적 조직에 관여하는 것으로 알려져 있습니다.
Rat과 Mouse에서 96%의 서열 동일성을 가지며, Human BASSOON 유전자는 클로닝 및 위치가 확인되었습니다.
Bassoon은 두 개의 double-zinc finger, 세 개의 coiled-coil 영역, 두 개의 polyglutamine domain을 포함하는 것으로 예측됩니다.
C-말단의 polyglutamine domain은 헌팅틴(Huntingtin)과 같은 단백질에서 신경퇴행성 질환과 관련이 있는 영역으로 주목받고 있습니다.
Bassoon은 시냅스 말단에서 postsynaptic density 반대편 부위에 집중되어 있으며, 배양된 뉴런에서 GABA(A) 및 GluR1 수용체와 공위치하는 것으로 확인되었습니다.
또한 presynaptic 단백질 Piccolo와도 공위치하며, 이는 신경전달물질 방출 부위에서 세포골격 조직화에 관여할 가능성을 시사합니다.
