MA1-4806 detects any kind of 6x-His tagged proteins in cells and complex cellular lysates.
It has been successfully used in flow cytometry, immunohistochemistry (frozen), immunofluorescence, Western blot, immunoprecipitation, and ELISA procedures.
By Western blot, this antibody detects recombinant proteins containing an epitope of at least six histidine residues, located at the N-terminus, C-terminus, or both termini.
A higher number of histidine residues leads to increased binding affinity of the antibody.
The MA1-4806 immunogen is recombinant (His)₆-p53 protein.

Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
It is commonly expressed at either N- or C-terminal regions of recombinant proteins to allow isolation or purification by immobilized metal affinity chromatography.
Epitope tagging enables detection of recombinant proteins for which no specific antibody is available.
His-tagged proteins are purified by metal chelate chromatography due to the metal-binding properties of histidine clusters.
For example, the affinity of the histidine-tag motif to Ni²⁺ is strong and selective enough to enable purification to homogeneity on a Ni²⁺-NTA adsorbent.
His tag-specific antibodies facilitate detection or co-immunoprecipitation of tagged proteins.
