Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10–20 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is tonsil or kidney tissue.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides.
It is expressed on cells of lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells in adult bone marrow, and cells from patients with chronic myelocytic leukemia (CML).
Additionally, CD10 is present on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on the brush border of kidney and gut epithelial cells.
CD10 cleaves peptides at the amino side of hydrophobic residues and inactivates peptide hormones such as glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin.
It is a 100 kDa type II transmembrane glycoprotein encoded by a gene with a 5′ untranslated region that is alternatively spliced into four mRNA transcripts without affecting the coding region.
Diseases associated with CD10 dysfunction include spinocerebellar ataxia 43 and Charcot-Marie-Tooth disease.
