Product Specific Information
The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity with other proteins has not been determined. Foxp3 (49–55 kDa) is a member of the forkhead/winged-helix transcriptional regulator family and is defective in scurfy (sf) mice. It is highly expressed in CD4⁺CD25⁺ regulatory T cells (Treg) and induces a Treg phenotype when expressed in CD4⁺CD25⁻ cells.
Immunoblotting with FJK-16s antibody maps the epitope to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced. Both isoforms are present in CD4⁺CD25⁺ lymphocytes, but not observed in mouse splenocytes, suggesting species differences in regulation.

Target Information
FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and expressed at high levels in CD25⁺CD4⁺ regulatory T cells. FOXP3 acts as a master regulator of Treg cells. Mutations in FOXP3 cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. Multiple transcript variants encoding different isoforms exist. In human breast and colon cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
