Product Specific Information
The monoclonal antibody 26mNOD2 reacts with mouse NOD2.
NOD2, also known as caspase recruitment domain-containing protein 15 (CARD15), is a cytosolic protein (~115 kDa) containing:
- A central NOD (nucleotide oligomerization domain)
- A C-terminal leucine-rich repeat (LRR) domain
- An N-terminal CARD domain
The LRR domain recognizes microbial components such as muramyl dipeptide, while the CARD domain interacts with RIP2 via homotypic CARD-CARD interaction, leading to downstream activation of NF-κB.
This antibody recognizes protein from transgenic and transfected cells.
Applications reported include immunoblotting (WB), and it has been validated on transfected cell lines and NOD2-transgenic mouse cells.
Target Information
NOD2 is a member of the apoptosis-regulating protein family that includes Apaf-1 and NOD1, containing:
- Two N-terminal CARDs
- A nucleotide binding domain (NBD)
- Multiple C-terminal leucine-rich repeats (LRRs)
NOD2 is primarily expressed in monocytes, whereas NOD1 is expressed in multiple tissues.
It regulates NF-κB signaling, acts as an intracellular receptor for bacterial lipopolysaccharides, and is associated with inflammatory bowel disease (IBD) and Crohn’s disease.
