- A suggested positive control for immunohistochemical applications is kidney from post streptococcal glomerulonephritis patients.
- Mouse anti-Human clusterin antibody, clone 3R3/2 recognizes clusterin, an ubiquitously expressed protein composed of two 40 kDa chains (alpha and beta) covalently joined by disulfide bonds.
Target Information
Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins. It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro).
Clusterin does not require ATP and maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70. It binds to cell surface receptors, triggering internalization and degradation of the chaperone-client complex.
Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis, while intracellular isoforms interact with ubiquitin and SCF E3 ubiquitin ligase complexes to promote proteasomal degradation of target proteins.
Additionally, clusterin modulates NF-kappa-B transcriptional activity, with nuclear isoforms promoting apoptosis and mitochondrial isoforms inhibiting BAX-dependent cytochrome c release.
