Target Information
BACE1 (beta-secretase)는 알츠하이머병(AD)에서 발견되는 아밀로이드 베타 펩타이드(Abeta)의 생성에 관여하는 주요 효소입니다. 일부 조기 발병 가족성 AD는 APP(amyloid precursor protein)의 ‘Swedish’ 변이에 의해 발생하며, 이는 BACE1에 의한 단백질 절단을 증가시킵니다. 이러한 연구 결과는 BACE1 및 기타 APP 절단 효소(예: gamma-secretase)의 억제를 목표로 하는 치료 접근법으로 이어졌습니다.
APP는 먼저 β-secretase에 의해 절단되어 용해성 단백질 유도체와 막 결합형 C99 단편을 생성합니다. C99는 이후 γ-secretase에 의해 절단되어 4 kDa 크기의 amyloid-β 펩타이드를 생성하며, 이는 알츠하이머병 환자의 뇌에 침착됩니다. β-secretase는 여러 연구 그룹에 의해 독립적으로 확인되었으며, beta-site APP cleaving enzyme (BACE) 또는 aspartyl protease 2 (Asp2)로 명명되었습니다. BACE/Asp2는 막관통형 아스파르틸 프로테아제로, APP와 공위치합니다.
