Target Information
BACE1 (beta-secretase)는 알츠하이머병(Alzheimer’s disease, AD)의 병리적 특징인 아밀로이드 베타 펩타이드(Aβ) 생성에 관여하는 주요 효소입니다.
일부 조기 발병형 가족성 AD는 APP(amyloid precursor protein)의 ‘Swedish’ 돌연변이에 의해 BACE1 절단이 증가하면서 발생합니다.
이러한 연구 결과는 BACE1 및 γ-secretase 억제제를 이용한 치료 전략 개발로 이어졌습니다.
APP는 먼저 β-secretase에 의해 절단되어 용해성 단백질 파생물과 막 결합형 99-아미노산 C-말단 단편(C99)을 형성하며, 이후 γ-secretase에 의해 4 kDa 아밀로이드-β 펩타이드가 생성되어 AD 환자의 뇌에 축적됩니다.
BACE1은 β-site APP 절단 효소로서, Asp2라고도 불리는 새로운 막관통형 아스파르틸 프로테아제로 APP와 공위치(colocalization)합니다.
