Target Information
Proteolytic degradation is critical for maintaining appropriate levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle, transcription, and signaling.
The ubiquitin-proteasome pathway deconstructs most proteins in the eukaryotic cytosol and nucleus, while others are degraded via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit (~31), barrel-shaped molecular machine (~2.5 MDa) composed of a 20S proteolytic core complex and one or two 19S regulatory subunits. The 19S subunits recognize ubiquitinated proteins and facilitate their unfolding and translocation into the 20S core.
The PA28/11S REG Activator complex (α, β, γ subunits) enhances 20S core activity. An enzymatic cascade attaches ubiquitin molecules to lysine residues of target proteins.
Defects in the ubiquitin-proteasome pathway are associated with genetic diseases such as cystic fibrosis (CFTR), Angelman’s syndrome (E6-AP), and Liddle syndrome (ENaC).
