Product Specific Information

Description:
The OX8 monoclonal antibody reacts with rat CD8 alpha, a member of the immunoglobulin superfamily. CD8α heterodimerizes with CD8β to form the CD8 antigen expressed on CD4+CD8+ thymocytes, most MHC class I-restricted T cells, and some macrophages. CD8 interacts with MHC class I on antigen-presenting cells and participates in T-cell receptor signal transduction via the kinase Lck.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.125 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction, T-cell differentiation, and activation. CD8 functions as a co-receptor with T-cell receptor (TCR) for MHC class I molecules, while CD4 serves as a co-receptor for MHC class II. CD8α is essential for MHC-I binding and is expressed on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production and cytotoxic T lymphocyte activation. Once activated, CTLs are critical for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
