Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimer or CD8αα homodimer. It is found on thymocytes, mature T cells, and NK cells. CD8 binds MHC class I and associates with p56lck to regulate T-cell development and activation.

Preliminary testing shows OKT8, RPA-T8, and HIT8a clones do not compete for binding to human leukocytes, indicating distinct epitopes.

Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.03 µg)/test (final volume 100 µL). Cell count range: 10⁵–10⁸ cells/test.

Target Information

CD8 is a type I transmembrane glycoprotein, acting as a co-receptor with TCR for MHC class I molecules. It exists as CD8αβ heterodimers or CD8αα homodimers on T cells, NK cells, monocytes, and dendritic cells. Binding to MHC-I/peptide complexes recruits Lck kinase, triggering CTL activation and pathogen clearance. Differentiation of CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
