Target Information

CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family.
It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and participates in receptor regulation via ubiquitin-mediated removal from the cell surface.
CD33 is expressed on cells of the myelomonocytic lineage and binds sialic acid residues in N- and O-glycans.
It serves as a therapeutic target for acute myeloid leukemia (AML) and is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells.
CD33 is also expressed on peripheral monocytes and is useful for distinguishing myelogenous leukemia from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
