Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha).
CD127 forms a complex with CD132 (common gamma chain) to create the functional IL-7 receptor (IL-7R).
It is a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways, including Jak/STAT.
IL-7 is essential for lymphocyte development and homeostatic proliferation of peripheral T cells.
IL-7R signaling also regulates TCR locus rearrangement in γδ T cells.

Recent studies show that CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs).
CD4⁺CD25⁺CD127⁻ cells are more highly enriched for Tregs, expressing Foxp3 and exhibiting suppressive activity in vitro.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells.
Use 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

CD127 (Interleukin-7 receptor, IL-7R) is a glycoprotein regulating lymphopoiesis.
The active receptor is an alpha/gamma chain heterodimer (IL7Rα/γc).
The γc chain activates signal transduction, while the α chain mediates specific signaling through cytoplasmic molecules.

CD127 supports proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells.
IL-7 binding to CD127 mediates critical immune functions.
Defects in CD127 are associated with severe combined immunodeficiency (SCID).
