Product Specific Information

For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation.
p53 activation leads to transcription of genes such as p21waf1, involved in G1 or G2/M arrest. Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerisation domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization. Ser392 phosphorylation is associated with tumor formation.
Mutations in the TP53 gene cause loss of tumor suppressor activity and are common in human cancers. Germline mutations also occur in cancer-prone families (e.g., Li-Fraumeni syndrome).
p53 plays roles in aging, oxidative stress, and learning/memory through effects on LTP.
