Product Specific Information

This CD25-4E3 monoclonal antibody reacts with human CD25, also known as the low-affinity interleukin (IL)-2 receptor alpha. CD25 is expressed on subsets of T cells (including regulatory T cells), activated T and B cells, and macrophages. It associates with CD122 (IL-2 receptor beta chain) and CD132 (common gamma chain) to form the high-affinity IL-2 receptor, involved in lymphocyte proliferation and survival. CD25 is a marker for Hodgkin’s disease and is associated with autoimmune diseases such as Type I diabetes.

This antibody is cross-reactive to rhesus monkey.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.06 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative).

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD25 (IL2RA) encodes the IL-2 receptor alpha chain, a cytokine receptor involved in T and B lymphocyte proliferation. The receptor complex includes IL2RA, IL2RB, and IL2RG, forming the high-affinity IL-2 receptor. Homodimeric IL2RA chains form low-affinity receptors; IL2RB homodimers form medium-affinity receptors. IL2 is produced by mature T cells and functions as a growth, differentiation, and apoptosis regulator. It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation. Mutations can cause interleukin 2 receptor alpha deficiency.
