Product Specific Information

Description:
The SN3 monoclonal antibody reacts with the human CD24 molecule (Heat Stable Antigen, HSA). This 35–50 kDa GPI-anchored molecule is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. P-selectin (CD62P) has been reported to bind CD24.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (100 µL final volume). Typical cell count: 10⁵–10⁸ cells/test.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the 488 nm blue laser. Recommended filter: 710/50 bandpass (alternative: 695/40 bandpass). Ensure instrument compatibility.

Fixation Recommendations:
Samples can be stored up to 3 days in the dark at 4°C using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)
  Minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development—from pro-B cells in bone marrow to mature surface Ig-positive B cells. Expression is low or absent in plasma cells. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity.
