Description:
Monoclonal antibody eBio4E3 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1). CD21 and CD35 are alternatively spliced transcripts from the Cr2 gene, producing proteins of 145 and 190 kDa respectively. These are expressed on mature B cells, but not on thymocytes, T cells, erythrocytes, or platelets. CD21 serves as a receptor for complement component C3d and Epstein-Barr virus (EBV), and participates in B-cell activation in association with CD19 and CD81. Cr2-deficient mice show impaired inflammatory and humoral immune responses.

The anti-mouse CD21/35 monoclonal antibody clones eBio4E3 and eBio8D9 bind different epitopes (no cross-blocking observed).

Applications Reported:

• Suitable for flow cytometric analysis.

Applications Tested:

• Tested on mouse splenocytes at ≤0.25 µg/test.
• A test is defined as the antibody amount required to stain a 100 µL cell sample.
• Recommended to titrate for optimal performance (10⁵–10⁸ cells/test).

Fluorochrome Information:

• PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
• Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
• Ensure instrument compatibility with this fluorochrome.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET compensation.
• Clone-specific fixation performance should be determined empirically.

Excitation / Emission / Laser:

• 488 nm / 710 nm / Blue Laser

Filtration:

• 0.2 µm post-manufacturing filtered.

Target Information

CD21 (Complement Receptor 2, CR2):
Binds C3 complement fragments, facilitating uptake and retention of immune complexes, survival of memory B cells, and humoral response development. It also serves as a receptor for Epstein-Barr virus and participates in prion targeting to follicular dendritic cells.

CD35 (Complement Receptor 1, CR1):
A membrane glycoprotein receptor for complement components C3b and C4b, regulating complement cascade activity. Found on blood cells, podocytes, and dendritic cells. Variations in CD35 form the basis of the Knops blood group system.
