Description:
The TR75-54 monoclonal antibody reacts with mouse Tumor Necrosis Factor Receptor II (TNFRII, TNFR-p80, TNFRSF1B, CD120b). TNFRII is expressed in various cell types and is strongly expressed on stimulated T and B lymphocytes. It is the main TNF receptor found on circulating T cells and mediates autoregulatory apoptosis in CD8+ cells. Soluble TNF receptors, truncated forms of the surface receptors, have neutralizing activity on both TNF-alpha and TNF-beta. TNF’s proinflammatory and immunosuppressive properties are believed to segregate at the receptor level, with soluble receptor-bound TNF acting as a reservoir to regulate TNF release.

Applications Reported:
This antibody has been reported for use in flow cytometry, immunoprecipitation, Western blot, and ELISA.

Applications Tested:
The TR75-54 antibody has been validated as a capture antibody in sandwich ELISA for mouse TNFRII detection when paired with the biotin-conjugated TR75-32 antibody (Product #13-1204) and recombinant mouse TNFRII as the standard. Recommended capture concentration is 1–4 µg/mL.
A standard curve using serial dilutions of recombinant standard (1000 pg/mL to 8 pg/mL) should be included in each ELISA plate.

Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

TNFR2 (tumor necrosis factor receptor superfamily member 1B) is a high-affinity receptor for TNFSF2/TNF-alpha and has lower affinity for TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. TNFR2 mediates most metabolic effects of TNF-alpha.
